You do not need to use both. Most often people use the Quietwalk Plus when over tile as it gives a little "forgiveness" that the Moisture Block doesn't .
Are the underlayment options either the black Moisture Block OR Quietwalk plus? Do we need both for a Marmoleum Click? We're putting it over tile. SAMPLE - Forbo Marmoleum Click Cinch Loc Seal, Cork Tree Waterproof Click - Floating
