Will this paint work on concrete that has had sealer applied. ?

It depends on what kind of sealer was applied. The concrete surface needs enough texture and porosity for the concrete paint to bond to it. If it is smooth and nonporous and you are determined to paint, you may need to take steps to acid etch or otherwise compromise the sealer.
