We are planning to install this under floating cork on top of an unsealed concrete basement floor. I understand this will protect the cork from moisture damage...however, is it possible that mold will grow between the plastic vapor barrier and the concrete? Thanks.

Yes, it is possible, although not very likely. At least it will not affect your flooring and will not expose anyone in the room. If you don't use a vapor barrier, the moisture emissions will be absorbed into the bottom of the floor and potentially cause cupping and damage your floor. The manufacturer will not warrant their flooring without it.
