Is there benefit to adding this under cork floor tiles in a second floor room on wood subfloor? Is it overkill?

Ultimately, you should always follow manufacturer instructions on the flooring you are installing. They will spell out what types of underlayment, and how thick you can go, for particular floors and subfloor combinations.
